The enzymatic oxidation of 1,2-propanediol phosphate to acetol phosphate.

چکیده
Lindberg (I) has presented evidence for the occurrence of 1,2propanediol phosphate in beef brain. This compound has also been reported to occur in other tissues (2-4). However, the structure, metabolism, and biological significance of this compound is unknown. Groth et al. (5) have reported that (Y-labeled pyruvate is incorporated into propanediol-P’ under anaerobic conditions in fluoride-inhibited homogenates of brain, liver, and induced tumors (Flexner-Jobling) . These authors have suggested that acetolphosphate is an intermediate in the conversion of pyruvate to propanediol-P. It has also been suggested that acetol-P is a product of the oxidation of propanediol-P by soluble a-glycerol phosphate dehydrogenase from rabbit muscle (‘3. In the present paper the product of the oxidation of propanediol-P by rabbit muscle oc-glycerol phosphate dehydrogenase has been established as acetol-P, CH3COCH20P03H2. Attempts to demonstrate this reaction in rat liver homogenates have been unsuccessful.

Formation in vivo of glycogen by certain intermediates of the lactate-propanediol pathway.
It has been observed by Miller and Olson (1) in experiments with heart muscle slices with the use of pyruvate and lactate as substrates in an anaerobic system that 0.1 M fluoride completely inhibited the utilization of pyruvate while lactate utilization was unimpaired. Since fluoride inhibits the enolase reaction in the Embden-Meyerhof glycolytic pathway, it was postulated that lactate could be...

متن کاملThe metabolism of acetone in rat.
Intraperitoneal injection of 5 mumol of acetone/g, body weight, into 3 rats previously fed 1% acetone (v/v) in their drinking water resulted in the appearance in blood serum of 16 +/- 2 nmol of 1,2-propanediol/ml and 8 +/- 1 nmol of 2,3-butanediol/ml. No detectable 1,2-propanediol or 2,3-butanediol was found in the serum of animals after acetone or saline injection without prior addition of ace...
متن کاملReduction of trioses by NADPH-dependent aldo-keto reductases. Aldose reductase, methylglyoxal, and diabetic complications.
The substrate specificities of human aldose reductase and aldehyde reductase toward trioses, triose phosphates, and related three-carbon aldehydes and ketones were evaluated. Both enzymes are able to catalyze the NADPH-dependent reduction of all of the substrates used. Aldose reductase shows more discrimination among substrates than does aldehyde reductase and is generally the more efficient ca...
متن کاملA microcolorimetric method for the determination of 1, 2-propanediol phosphate.
Recent interest in the metabolism of 1,2-propanediol phosphate (l-9) has made desirable the development of a specific assay procedure for the determination of this compound in tissues. The extreme stability of the phosphorylated compound to strong acid and to alkali (1, 10) and its ready hydrolysis to 1,2-propanediol and phosphate by both acid and alkaline phosphatase (1) have led to the develo...
